Architecture of organic farms in Czech republic

New agricultural constructions in Czech Republic happen, in better case, privilege of civil engineer, in the worse case only privilege of the investor, in the past. While architects or enlightenment project architects with inventive and feeling for architecture, rarely interfered to their projection. In the practice, the preferred and implemented misconception, that it is a purely purpose-built buildings with economical advantageous solution type, when external design of the building and his effect in landscape plays just a minor role, meant a huge mistake, which disfeature face of a lot of villages. With the gradual development of organic and sustainable agriculture is obvious, that to the design of these buildings gets back the idea of invention and emotion of construction into the landscape. Essence of a dissertation is how to proceed with the draft of a new or completion of original organic farm in conditions of the Czech Republic and on which specific points to focus in the design. Objective of thesis is generalization of rules design new modern organic farm or new buildings on the existing organic farm, using the principles of organic architecture and architecture undemanding of energy, renewable source of energy and non-waste management and verification on the specific examples in the Czech Republic and neighboring Europa countries. Where else should apply the principles of organic architecture more than in the construction of organic farms. Moreover, organic farm offers a great number of functions and with related different operations with requirements for specific areas. This is an added bonus, diversity of construction and the possibility of building creativity in architectural solutions. It is then mainly limited functionality of the building, legislative, public health, fire and traffic regulations, with which any proposal must cope. Organic farms are a phenomenon, but also the future of sustainable agriculture in the Czech Republic, as well. Not only production side of organic farms, but also neglected building stock deserves the attention of experts. Only organic farm with the right comprehensive created building and energy concept on an ecological principle is becoming a well-functioning self-contained unit connected with the cycle of nature, the surrounding landscape and regional tradition.
Application of Mass Spectrometry for the Determination of Oxidative Stress Markers and Mycotoxins

The first topic presented in the dissertation thesis is determination of isoprostanes as markers of oxidative stress and other compounds affected by presence of oxidative stress. Isoprostanes iPF2-III, iPF2-VI, iPF2-VI, astaxanthin and polyunsaturated fatty acids (PUFA), especially arachidonic acid (AA) were monitored in Atlantic salmon eggs (Salmo salar). Methods for the determination of these compounds have been developed and optimized using chromatographic separation coupled to conventional or mass spectrometric detection. Freshly laid eggs, eyed embryos and non-viable eggs were used to test a general hypothesis that egg viability can be affected by susceptibility to oxidative stress, either through the specific fatty acid concentration and/or the antioxidant capacity of the eggs. Levels of isoprostanes and arachidonic acid (AA) were significantly higher in non-viable eggs than in control (eyed embryos) as well as relative abundance of PUFA. While no difference of isoprostanes was found between freshly laid and control those from the Atlantic stock except iPF2-VI which was observed under the LOQ in the control. Higher levels of PUFA and AA in comparison with the control were observed in the freshly laid eggs. However, the only statistically significant difference was observed in the amount of astaxanthin. Different levels of PUFA and astaxanthin may be related to their biochemical consumption during the development of eggs. This work evaluated potential effect on the viability of eggs Salmo salar due to the presence of oxidative stress. The monitoring of mycotoxins in food and feed was the subject of the second topic. Mycotoxins are secondary metabolites produced by fungi. They are ubiquitous undesirable natural contaminants that are toxic for humans and animals. Today are known more than 500 mycotoxins. However, only few of them are regulated by the European Union. The European Food Safety Authority (EFSA) was asked by the European Commission to provide a scientific opinion on other mycotoxins for which statutory limits could be developed. In this study is proposed simultaneous screening allowing fast, reliable and sensitive approach, identification and quantification of 17 mycotoxins in food and feed sample. The method includes both mycotoxins regulated by the EU and selected mycotoxins required by the EFSA (aflatoxins, deoxynivalenol, nivalenol, zearalenone, fumonisin, ochratoxin A, T-2 toxin, HT-2 toxin, enniatins and beauvericin). Analytes are isolated by the modified QuEChERS method. For separation and target mycotoxins detection, ultra-performance liquid chromatography coupled to tandem mass spectrometry (UPLC –MS/MS) was employed. The method also allows determination of ergot alkaloids (ergocornine, ergosine, ergocryptine, ergocristine and their respective epimers). The developed method was used either for monitoring mycotoxins and ergot alkaloids in feed and raw materials and barley and malt prepared from it.
Comparison of quality of milk from conventional and organic farming

The diploma thesis focuses on the comparison of milk quality in two different ways of farming, conventional and organic farming. Almost a hundred parameters on the set of 64 bulk milk samples were monitored. The theoretical part of the paper describes the composition of cow's milk, its technological and hygienic properties and it notes the nutritional impact of milk and milk products. It also mentions the most important legal definition of organic farming. Experimental part describes the setting of the experiment and mentions the methods used to analyze milk samples for individual parameters and it details the ten selected determination. The difference in measured values of various milk indicators depending on the dairy farm was observed by using statistical methods and an explanation was suggested in the case of the statistical significance. The thesis was based on the cooperation of the Faculty of Chemistry VUT Brno and a National reference laboratory for raw milk at the Research Institute for Cattle Breeding in Rapotin.
The Study and Performance of Organic Farms Increased

This thesis deals with performance of a company Horňácká Farma s.r.o. processing and marketing organic beef meat. The goal is to find ways to increase the performance of the farm. Based on the analysis of the current state recommendations to remove bottlenecks in the process have been made. These recommendations generally contribute to higher business performance.
Urban and architectural study of environmentally friendly tourist area Strachotín.

ANNOTATION On the banks of the Střední Novomlýnská reservoir, in the cadastre of the municipality of Strachotín, the diploma thesis presents the overall design of a multifunctional complex, based on a sensitive connection between organic farming, well-breeding, and eco-tourism. The proposal shows the concept of farming in a "closed circle", with regard to the environment. The urban solution deals with the selected land with an area of 20 ha. The architectural part closely introduces the system of objects, creating a new landscape, perceived by both a human and bird's eye perspective. The layout solution analyzes in detail the functional arrangement of the farm, trying to meet all the requirements for efficient management while maintaining the organic regime and the coexistence of visitors and employees. The complex also offers a restaurant, multifunctional hall, conference rooms, classrooms, workshops, a gym, and exhibition areas. Accommodation is provided by a system of separate buildings, presenting the current possibilities of natural construction and environmentally friendly technologies to the guests, while maintaining the comfort of living. The farm is fully equipped for weddings or corporate events. The design and material solution use a maximum of natural materials in combination with current technologies. In addition to designing a viable multifunctional unit, the aim of the thesis is also to bring added value to the region, by holding annual cultural events and regular activities for children and seniors from the surrounding municipalities. At the same time, the work aims to create a sensitive counterpoint to current tourist destinations in the area.

The bachelor thesis describes organic farming in general terms, explains the function of certification organisations and compares the basic criteria. It focuses on the differences in subsidy policy in the compared countries, which are the Czech Republic, Slovakia and Austria. It also seeks to highlight the possibilities and opportunities in the field of organic farming. It also highlights the weaknesses and strengths of the sector. It finally suggests possible further developments in organic farming.
Porovnání výnosu a kvality zrna u vybraných odrůd pšenice ozimé v konvenčním a ekologickém způsobu pěstování

This bachelor thesis aimed to compare the yield and grain quality of eight varieties of winter wheat grown using conventional and organic cultivation practice. The experiment was conducted at the Field Experimental Station in Žabčice, and the varieties included Genius, Butterfly, LG Orlice, Illusion, Pirueta, Mercedes, LG Absalon, and RGT Telemark. All these varieties were registered for conventional farming and are tested for suitability for organic farming. Two experiments were established in the same field with the same fore crop of alfalfa (Medicago sativa). The organic experiment was grown in accordance with eco-farming regulations. The basic parameters for baking quality were determined at the Breeding Station RAGT Czech s.r.o. in Branišovice. This thesis compares results of yield, thousand seed weight, protein concentration in dry matter, Zeleny sediment test, falling number amount and wet gluten content. Yield of most of varieties was higher in the conventional experiment than in the organic experiment. The average yield of the conventional experiment was 0,56 t·ha-1 higher than the average yield of the organic experiment. The difference of thousand seed weight and all of tested parametres of quality ended same as in yield comparison. The conventional experiment was 0,5 g higher in thousand seed weight than the organic experiment. In Zeleny sediment test was results of conventional experiment 2,5 ml higher than in the organic experiment. The conventional experiment had 0,6 % higher protein concentration in dry matter. The conventional experiment had 26 s higher falling number than the organic experiment. The wet gluten content was 0,7 % higher in conventional experiment than in organic experiment. Overall the conventional experiment had higher yield and better quality in all aspects than the organic experiment.
Vliv obnovy travních porostů na produkci a kvalitu píce a na rentabilitu u ekologicky hospodařícího podniku

The diploma thesis evaluates the effects of different methods of grasslands renovation. Three variants were compared – original permanent grassland, renovation using commercial mixture and renovation with regional mixture, at two sites in the village of Štítná nad Vláří – Popov in the White Carpathians. The literary research is dedicated to the importance of grasslands and their restoration in accordance with the principles of ecological farming. Furthermore, the conditions of the White Carpathians Protected Landscape Area are presented. The research part describes the results of the annual yield of dry fodder, the quality of the grassland according to the feed value and the representation of plant species. The stand renewed with the commercial mixture showed the highest yields (3.73 t/ha) and quality, with the regional mixture the effect of renewal was not evident. At the end, an economic evaluation of the renewal of the grassland is carried out using a contribution margin. It was minus 2093 CZK/ha for the given trial, the yield threshold was set at 4.7 t/ha. Restoration using a regional mixture was evaluated as too costly.

This thesis examines the impact of sustainable food production on consumer behaviour. In order to achieve the aim of the thesis, an analysis of organic farming in the EU and the Czech Republic, an analysis of the organic food market in the Czech Republic and an assessment of the willingness to pay a higher price for or-ganic food were carried out. A questionnaire survey was also conducted among consumers to determine their opinion on organic products and their purchasing behaviour towards these products. As a result of the thesis, it was determined which factors influence consumers' purchasing behaviour towards organic products and which factors are barriers to the purchase of organic products. Recommendations for consumers and producers involved in the production and sale of organic food were also formulated.
The Voluntary Tools of Environmental Protection

The diploma thesis is focused on the voluntary tools of environmental protection, those representing a remarkably wide and variable group of tools. The aim of this thesis is to create an integrated and compendious overview of the previously mentioned tools applied not only in the Czech Republic, to outline its basic characteristics, ways of its usage, and its mutual combinations, including the analysis of the legal regulation of the voluntary tools. After the two introductory chapters dedicated to the environmental policy and the system of all tools of environmental protection the following chapter deals with the voluntary tools only, dividing them for the purposes of this diploma thesis into categories of regulatory, informative, and educational tools. Considering the extent of the topic, the author decided on a detailed analysis elaboration of the three chosen tools in the following part of the thesis. In the fourth chapter the diploma theses concentrates on the EMAS according to the Regulation (EC) No 1221/2009 of the European Parliament and of the Council on the voluntary participation by organisations in a Community Eco-Management and Audit Scheme (EMAS), which is being compared to the second implementation option of Environmental Management System, that is represented by the internationally...
